Development of Monoclonal Antibodies for Diagnosis of Plasmodium vivax
نویسندگان
چکیده
Plasmodium lactate dehydrogenase (pLDH) is a strong target antigen for the determination of infection with Plasmodium species specifically. However, a more effective antibody is needed because of the low sensitivity of the current antibody in many immunological diagnostic assays. In this study, recombinant Plasmodium vivax LDH (PvLDH) was experimentally constructed and expressed as a native antigen to develop an effective P. vivax-specific monoclonal antibody (mAb). Two mAbs (2CF5 and 1G10) were tested using ELISA and immunofluorescence assays (IFA), as both demonstrated reactivity against pLDH antigen. Of the 2 antibodies, 2CF5 was not able to detect P. falciparum, suggesting that it might possess P. vivax-specificity. The detection limit for a pair of 2 mAbs-linked sandwich ELISA was 31.3 ng/ml of the recombinant antigen. The P. vivax-specific performance of mAbs-linked ELISA was confirmed by in vitro-cultured P. falciparum and P. vivax-infected patient blood samples. In conclusion, the 2 new antibodies possessed the potential to detect P. vivax and will be useful in immunoassay.

Development of Monoclonal Antibodies That Target 1-Cys Peroxiredoxin and Differentiate Plasmodium falciparum from P. vivax and P. knowlesi
Prompt and accurate diagnosis of malarial patients is a crucial factor in controlling the morbidity and mortality of the disease. Effective treatment decisions require a correct diagnosis among mixed-species malarial patients. Differential diagnosis is particularly important in cases of Plasmodium vivax, a species that shares endemicity with P. falciparum in most endemic areas. Moreover, it is ...
متن کاملDevelopment and Characterization of Monoclonal Antibodies against Erythrocytic Stages of Plasmodium Vivax and Use in Detection of Malaria
Hybridomas producing monoclonal antibodies (MAbs) directed against P. vivax erythrocytic stage antigens were isolated. Here, we describe the production of six MAbs. Culture supernatant from growing hybridomas was tested for their reactivity with P. vivax erythrocytic stages by ELISA against P. vivax crude (PvC) antigen and by immunofluorescence assay on P. vivax infected erythrocyte smears. P. ...
متن کاملOptimized Method for Purification of Expressed Plasmodium Vivax Duffy Binding Protein-II (PvDBP-II): Implication for Vivax Malaria Vaccine Development
Background: The purity and correct folding of a recombinant protein is critical for any structural, biochemical and vaccine design studies. Plasmodium vivax Duffy binding protein-II is a leading vaccine candidate for vivax malaria. In the present study, the purification process of recombinant DBP-IX (a variant form of PvDBP-II) was optimized to achieve the highest yield and purity. Moreover, ...

Objective(s): Leishmaniasis is endemic in 88 countries. Amastigote forms of Leishmania are experts at exploiting host cell processes to establish infection. Monoclonal antibodies are key reagents used in the diagnosis of infectious and non-infectious diseases. The aim of this study was to produce monoclonal antibodies against axenic amastigotes of the Leishmania infantum strain in Iran.Material...
متن کاملCircumsporozoite proteins of human malaria parasites Plasmodium falciparum and Plasmodium vivax
Monoclonal antibodies were raised against sporozoites of two species of malaria parasites, Plasmodium falciparum and Plasmodium vivax. The antibodies reacted with polypeptides (circumsporozoite proteins) that are uniformly distributed over the entire surface of sporozoites, as shown by indirect immunofluorescence and by the circumsporozoite precipitin reaction. The epitopes recognized by the mo...
